Is flow control (RTS/CTS) mandatory?
I only use TX RX to download ,but failed.

No RTS/CTS is not mandatory, and what exactly you were trying to do and failed, download code to the sysram or to the flash ? Also since you want to donwload from UART you will need the reset, in order for the bootloader to start running (as the Smart Snippets Toolbox indicates).
